Afatinib figure - C797S

Hi, Your figure "Afatinib covalently binds to cysteine number 797 of the epidermal growth factor receptor (EGFR) via a Michael addition (IC50 = 0.5 nM).[12]" is extremely interesting. My wife has NSCLC for 7 years, but has recently progressed on Tagrisso with C797S, retaining T790M.

Afatinib appears to offer a benefit in reacting with C797S, however short lived.

Could you provide a reference for the Afatinib / C797S interaction ? [I am trying to get Schubert-Zsilavecz, M, Wurglics, M, Neue Arzneimittel Frühjahr 2013.]

Hi! Schubert-Zsilavecz is a seminar script and unlikely to be found on the internet. I have found a description of this mechanism here. The C797S mutation means there is no cysteine at position 797 but a serine, so afatinib probably won't be able to bind. This looks like a source for this assumption. However, that's all preclinical data that won't tell you whether and for how long your wife may benefit from afatinib. Sorry I can't help you more. I have emailed the two gentlemen for the paper and the latest state of play. Meanwhile, I found "https://www.jto.org/article/S1556-0864(19)30599-4/fulltext" which indicates the problem of treating C797S is unsolved.

Removal of link

hello there, Hope this message greets you well. Thanks for letting me know. I placed the link because there were no links or page related to Vraylar medicine on Wikipedia. And this post had the mechanism of action and the I just went through the guidelines now. It says the external links can be linked in the reference section for further read if it has more information regarding the topic. And the post I linked has a detailed explanation on the mechanism of action, side effects, written by doctors. I thought this can be helpful. @Poseidon3211: Hi! healthcondition.org seems to be a private website; this does not meet the standard requited by medical articles on Wikipedia as far as I can tell. I have linked to the official drug information on drugs.com. Ideally, of course, the information would be in the Wikipedia article itself :-) See WP:MEDRS for how to identify reliable sources for medical articles. The true and forgotten discoverers of rapamycin.

My name is Gionei Gomes da Silva, and I am a Brazilian physician. All publications on the discovery of rapamycin forget that the real discoverers were the Brazilians​. It was really Canada that motivated the Brazilian researchers to go to Rapa Nui Island. It is also true that the researcher Suren N. Sehgal was responsible for the discovery of the magnificent properties of rapamycin. However, if the Brazilians had not discovered it the world would not have known it! I never could identify the names of the Brazilians. Because the commemorative plaque placed on Rapa Nui Island, in 2000, by Wyeth, in honor of the Brazilian scientists, does not contain their names.
